A Comparative Clinical Study on the Antidepressant Effect of Lofepramine




Abstract
In order to evaluate therapeupic efficacy and side effects of lofepramine, double-blind trial of lofepramine and amitriptyline were conducted.
The subject were 42 consecutive psychiatric inpatients or out-patients who had the diagnosis of depression during the period from Sept. 1, 1984 to Feb. 28, 1985.
The results were as follows:
1. Comparing with amitriptyline and lofepramine, they showed equally potential antidepressant effects as measured by Hamilton¢¥s depression scale (HAM-D) and lung¢¥s self-rating depression scale(SDS).
2. Global improvement rate assessed by patients and doctors demonstrated that both drugs were clinically effective in terms of symptomatic improvement, however, no difference was noted between lofepramine and amitriptyline.
3. There was significant difference between two drugs on side effect measured by DOTES (Dosage Record and Treatment Emergent Symptom Scale), lofepramine group revealed fewer side effects including anticholinergic symptoms than amitriptyline group.
